1. What Heavy Metal Testing Is
Heavy metal testing measures toxic elemental contamination in a peptide batch. Common metals tested include:
- Lead (Pb)
- Arsenic (As)
- Cadmium (Cd)
- Mercury (Hg) Some labs also screen for nickel, chromium, cobalt, aluminum, and copper if relevant.
2. Why It Matters for Peptides
- Heavy metals can be introduced from:
- Contaminated raw materials.
- Poor-quality glassware or reaction vessels.
- Impure solvents and reagents.
- In regulated pharma, metal levels are strictly limited under ICH Q3D guidelines.
- Even in “research use only” markets, high heavy metal content can indicate unsafe manufacturing practices.
3. Common Testing Methods
A. ICP-MS (Inductively Coupled Plasma Mass Spectrometry) – Most Common
- The gold standard for ultra-trace detection (parts per billion / ppb).
- Works by:
- Dissolving peptide sample.
- Atomizing it in a plasma torch (~10,000 °C).
- Measuring each element’s unique mass-to-charge ratio in the MS detector.
Pros: Extremely sensitive, covers a wide range of metals. Cons: Requires specialized lab equipment.
B. ICP-OES (Inductively Coupled Plasma Optical Emission Spectroscopy)
- Similar to ICP-MS, but detects light emitted from excited atoms instead of mass.
- Slightly less sensitive than ICP-MS but still highly reliable.
C. AAS (Atomic Absorption Spectroscopy)
- Older method using light absorption by vaporized atoms.
- Cheaper, but usually tests only one metal at a time.
4. Interpreting Results
Results are typically given in:
- µg/g (ppm) or ng/g (ppb).
- Compared to limits set by:
- ICH Q3D (Pharmaceutical standards).
- USP <232> Elemental Impurities guidelines.
Example limits for injectables (ICH Q3D, parenteral):
- Lead: ≤0.5 µg/g
- Arsenic: ≤1.5 µg/g
- Cadmium: ≤0.2 µg/g
- Mercury: ≤0.3 µg/g
A high-quality peptide should be well below these limits.
