1. What Heavy Metal Testing Is

Heavy metal testing measures toxic elemental contamination in a peptide batch. Common metals tested include:

  • Lead (Pb)
  • Arsenic (As)
  • Cadmium (Cd)
  • Mercury (Hg) Some labs also screen for nickel, chromium, cobalt, aluminum, and copper if relevant.

2. Why It Matters for Peptides

  • Heavy metals can be introduced from:
    • Contaminated raw materials.
    • Poor-quality glassware or reaction vessels.
    • Impure solvents and reagents.
  • In regulated pharma, metal levels are strictly limited under ICH Q3D guidelines.
  • Even in “research use only” markets, high heavy metal content can indicate unsafe manufacturing practices.

3. Common Testing Methods

A. ICP-MS (Inductively Coupled Plasma Mass Spectrometry)Most Common

  • The gold standard for ultra-trace detection (parts per billion / ppb).
  • Works by:
    1. Dissolving peptide sample.
    2. Atomizing it in a plasma torch (~10,000 °C).
    3. Measuring each element’s unique mass-to-charge ratio in the MS detector.

Pros: Extremely sensitive, covers a wide range of metals. Cons: Requires specialized lab equipment.


B. ICP-OES (Inductively Coupled Plasma Optical Emission Spectroscopy)

  • Similar to ICP-MS, but detects light emitted from excited atoms instead of mass.
  • Slightly less sensitive than ICP-MS but still highly reliable.

C. AAS (Atomic Absorption Spectroscopy)

  • Older method using light absorption by vaporized atoms.
  • Cheaper, but usually tests only one metal at a time.

4. Interpreting Results

Results are typically given in:

  • µg/g (ppm) or ng/g (ppb).
  • Compared to limits set by:
    • ICH Q3D (Pharmaceutical standards).
    • USP <232> Elemental Impurities guidelines.

Example limits for injectables (ICH Q3D, parenteral):

  • Lead: ≤0.5 µg/g
  • Arsenic: ≤1.5 µg/g
  • Cadmium: ≤0.2 µg/g
  • Mercury: ≤0.3 µg/g

A high-quality peptide should be well below these limits.
