Reconstituting a lyophilized (freeze-dried) peptide is a process that requires a sterile environment and careful handling to maintain the integrity and potency of the compound. Bacteriostatic water (BAC water), such as the Hospira brand, is the recommended solvent because it contains 0.9% benzyl alcohol, which inhibits the growth of bacteria, allowing the reconstituted solution to be stored for a longer period in the refrigerator. Here are the step-by-step directions on how to reconstitute a vial of peptide using Hospira bacteriostatic water:

Preparation and Safety

  1. Calculate the Volume: Determine the volume of bacteriostatic water needed to achieve your desired concentration (e.g., to create a 1 mg/mL solution from a 10 mg peptide vial, you would need 10 mL of water). You may use an online peptide reconstitution calculator for accuracy.
  2. Gather Supplies: Ensure you have the lyophilized peptide vial, the Hospira Bacteriostatic Water vial, sterile syringes (typically a 3 mL or 5 mL syringe for mixing and an insulin syringe for drawing the final product), and several alcohol prep pads.
  3. Clean Your Workspace: Thoroughly wash your hands with soap and water and put on gloves. Clean a flat, sterile work surface and arrange all your supplies.
  4. Equalize Temperature: If the peptide vial was stored in the refrigerator or freezer, allow it to sit at room temperature for 10-15 minutes before reconstitution.

The Reconstitution Process

  1. Clean the Vials: Flip the caps off both the peptide vial and the Hospira Bacteriostatic Water vial. Use a new alcohol prep pad to thoroughly swab the rubber stopper of each vial and allow them to air dry completely for 30-60 seconds. Do not blow on them or wipe them with a towel.
  2. Draw Air and Water: Take your large mixing syringe and pull the plunger back to draw air into the barrel equal to the volume of bacteriostatic water you plan to use (the amount you calculated in Step 1).
    • Insert the needle through the center of the rubber stopper on the bacteriostatic water vial.
    • Inject the air into the vial (this helps create positive pressure).
    • Invert the vial and slowly draw the calculated volume of water into the syringe.
  3. Inject into Peptide Vial: Locate the peptide vial. Carefully insert the needle through the rubber stopper.
    • Crucial Step: Inject the bacteriostatic water slowly, aiming the needle tip so that the water gently flows down the inside wall of the peptide vial. Do not spray the water directly onto the powder, as this can damage the peptide’s delicate structure.
  4. Mix the Solution: Once all the water has been added, remove the mixing syringe and immediately dispose of it in a sharps container.
    • Do not shake the peptide vial vigorously.
    • Gently swirl the vial in slow, circular motions or lightly roll it between your palms for 30-60 seconds.
    • Allow the vial to sit for a few minutes. The lyophilized powder should completely dissolve, leaving a clear, homogeneous liquid solution. If particles remain, repeat the gentle swirling/rolling until clear.

Storage and Final Steps

  1. Label and Store: Once fully reconstituted, immediately place the peptide vial into the refrigerator ($2^{\circ}$C to $8^{\circ}$C).
    • Label the vial with the date of reconstitution and the new concentration (e.g., “10mg/mL - Reconstituted ”).
  2. Safety: Always store the peptide away from light and excessive heat, and discard the reconstituted solution according to the compound’s specific shelf-life guidelines.
