Sterility testing confirms that a peptide preparation — typically a lyophilized vial — is free from viable microorganisms like bacteria, yeast, and mold. Even in “research use only” settings, this test is an important quality benchmark.


1. Why Sterility Testing Matters

  • Verifies that no living contaminants are present.
  • Confirms proper aseptic handling, lyophilization, and packaging.
  • Adds trust and credibility in light gray market or group-buy settings.
  • Essential for anything intended for parenteral (injectable) use.

2. Testing Methods

A. USP <71> Membrane Filtration (gold standard for clear solutions)

  • Reconstitute sample and pass it through a sterile 0.45 µm filter.
  • Filter is placed in:
    • Fluid Thioglycollate Medium (FTM) – detects anaerobic bacteria.
    • Soybean-Casein Digest Medium (SCDM) – detects aerobic bacteria and fungi.
  • Incubate for 14 days at two different temperatures.
  • No growth → Pass.

B. Direct Inoculation

  • A reconstituted sample is added directly into both media.
  • Incubate for 14 days under aerobic and anaerobic conditions.
  • Observe for turbidity or visible microbial growth.

3. Optional Add-On Tests

  • Endotoxin Test (LAL Assay) – Detects bacterial endotoxins.
  • Particulate Matter – Checks for visible and microscopic particulates.
  • pH & Osmolarity – Confirms safe reconstitution parameters.

4. Sample Requirements

  • Usually 1 vials per batch.
  • Reconstitute in sterile bacteriostatic or sterile water before submission.
  • Handle in a cleanroom or biosafety cabinet to avoid false positives.

5. Timeline & Cost

  • Incubation: 14 days (per USP standards).
  • Total turnaround: ~15–18 days.
  • Cost: $150–$350 for sterility
